Mechanism of lifetime determination of ubiquitinated proteins by selective autophagy

Selective autophagy contributes to the regulation of “protein lifespan” in cells, and ubiquitinated proteins are known as representative substrates. However, what kind of ubiquitinated proteins are degraded by selective autophagy, by what mechanism, and for what purpose, has not yet been fully elucidated. In this study, we will comprehensively identify ubiquitinated proteins that are degraded by selective autophagy using an original and innovative substrate purification method, and elucidate the mechanism of degradation of these substrates. Furthermore, using vertebrate models, we will elucidate the physiological significance of protein lifespan regulation by selective autophagy. This research is expected to advance our comprehensive understanding of the mechanisms and significance of protein lifespan regulation.
